CHROMOSOME NUMBER, MEIOTIC BEHAVIOR AND POLLEN VIABILITY IN THREE SPECIES OF STYLOSANTHES SW. (LEGUMINOSAE-PAPILIONOIDEAE) NATIVE TO SOUTHERN BRAZIL

Abstract

In this paper ten accessions of three species of leguminous forage Stylosanthes Sw. were analyzed with regard to the recount of the number of chromosomes, meiotic behavior and the percentage pollen viability by comparing three different dyes. In order to count the chromosome number, the meristematic regions of the roots were treated with pectinase-celullase 2% and stained with propionic orcein 2%. In the meiosis study and the estimation of pollen viability the flower-buds were fixed in absolute ethanol: chloroform: acetic acid (6:3:1). The material for meiosis was stained with lactopropionic orcein 1% and the pollen was stained with acetic orcein 1%, Alexander reactive and propionic carmine 1%. All accessions were diploids with 2n=2x=20 chromosomes. The meiotic behavior concerning microsporogenesis was considered normal with the meiotic index (MI) higher than 90%. In the estimation of pollen viability of these species the most dependable pigment was the Alexander reactive because it could differentiate through colors the cellular wall and the protoplasm.
