Crioconservação do sêmen da garoupa-verdadeira Epinephelus marginatus

Resumo

Este trabalho teve a finalidade de desenvolver um protocolo de crioconservação do sêmen da garoupa-verdadeira Epinephelus marginatus. Em três experimentos foram analisados os efeitos de três diluentes (pH 6,1; 7,8 e 8,2), quatro diluições (1:0; 1:1; 1:2 e 1:3), seis concentrações de crioprotetor dimetilsulfóxido (0; 2,5; 5,0; 7,5; 10,0 e 12,5%) e cinco velocidades de congelamento (90, 60, 45, 30 e 15°C.min-1) sobre a motilidade e o tempo de motilidade espermáticas no sêmen crioconservado. O sêmen foi congelado em vapor de nitrogênio empregando-se palhetas criogênicas, e posteriormente mantido em nitrogênio líquido. O tratamento que propiciou maior motilidade e tempo de motilidade espermáticas (p<0,05) foi aquele proporcionado pelo emprego do diluente B (pH 7,8), na proporção de 1:3 (v/v), com crioprotetor (dimetilsulfóxido) a 5% e em uma velocidade de congelamento de 60°C.min-1. Estes resultados possibilitaram a implantação do primeiro banco de sêmen da garoupa-verdadeira no Brasil.
